Resumo: A phylogenetic analysis of the Neoplecostominae was performed under the maximum parsimony optimality criterion, based on characters originally proposed in this study as well as on characters from previous analyses. Seventy one taxa were included in the analysis, 12 as outgroup and 59 as ingroup. In this analysis all species of Pareiorhaphis, Pareiorhina and Kronichthys were included as well as most of the species of Neoplecostomus and Isbrueckerichthys. Were also included five genera of the Hypoptopomatinae (Parotocinclus, Eurycheilichthys, Epactionotus, Otocinclus and Hisonotus), all species of Hemipsilichthys and most of Delturus. The analysis of the 303 characters from external morphology and anatomy resulted in two maximally parsimonious cladograms with 2171 steps, consistency index of 0. 21 retention index of 0. 66. The genera Neoplecostomus, Isbrueckerichthys, Pareiorhaphis, Pareiorhina, and Kronichthys were recovered as monophyletic. The subfamily Neoplecostominae is considered monophyletic and includes the genera Neoplecostomus, Isbrueckerichthys, Pareiorhaphis and an undescribed genus. The relationships among the genera included in the Neoplecostominae recovered Isbrueckerichthys and Neoplecostomus as sister-groups and more related to Pareiorhaphis. The new genus is basal, sister to the genera mentioned above. Pareiorhina and Kronichthys were recovered as sister to each other and more related to the members of the Hypoptopomatinae, and are transferred to that subfamily. The basal position of Delturus and Hemipsilichthys in the family was confirmed in the present analysis, corroborating previous hypothesis. Delturinae includes the genera Delturus and Hemipsilichthys. Phylogenetic relationships among Delturinae are (Hemipsilichthys nimius (H. gobio + H. papillatus)) (Delturus brevis (D. angulicauda + D. parahybae)).Several new species were recognized: six Pareiorhaphis, one Neoplecostomus, two Pareiorhina, one Kronichthys, and one Hisonotus. Finally, three new genera were recognized, one Neoplecostominae and two Hypoptopomatinae.
