They represent waste disposal, or they are used for wine alcohol production, as source of grapeseed oil, fertilizer, or for animal feed. However, these by-products are important sources of nutraceutical molecules with functional properties, including polyphenols with antioxidant activity. This study aimed to valorize these cellulosic materials from winemaking as source of bioactive phenolic compounds and to produce oligosaccharides with prebiotic activity. Grapes from Touriga Nacional and Marselan varieties (Vitis vinifera L.) were used for conventional red winemaking in the winery pilot plant of the Instituto Superior de Agronomia. The summative analysis was carried out to determine the chemical composition of the grape pomace (skins and seeds) and stems of both varieties: ash content, organic content as extractives (by sequential extraction in Soxhlet using dichloromethane, ethanol, and water), lignin fraction, and structural polysaccharides (cellulose and hemicelluloses). The amount of extractives was high and around 64.1% (d.w.) for both pomaces, and 46.2% and 59.5% for Marselan and Touriga Nacional stems. The main contribution came from polar compounds solubilized by ethanol and water, representing 82% of the total extractives, with total phenols contents ranging from 213.4 in Touriga Nacional stems to 457.1 g GAE/kg of dry material in Marselan pomaces. After extractives removal, the pomaces and stems were submitted to a hydrothermal treatment to get oligosaccharides from hemicelluloses. The autohydrolysis was carried out following a central composite rotatable design as a function of temperature (142-198°C) and time (48-132 min). For the materials of both varieties, the production of sugars by autohydrolysis showed to increase with reaction temperature and time. The highest sugar yields varied from 116.3 to 168.3 g of glucose/kg dry biomass.
